CD160 is a 27 kDa glycoprotein, also known as BY55, NK1, or NK28, which was initially identified with the monoclonal antibody BY55. Its expression is tightly associated with peripheral blood NK cells and CD8 T lymphocytes with cytolytic effector activity. The cDNA sequence predicts a cysteine-rich, glycosylphosphatidylinositol-anchored protein of 181 amino acids with a single Ig-like domain weakly homologous to KIR2DL4 molecule. CD160 is expressed at the cell surface as a tightly disulfide-linked multimer. It has both stimulatory and inhibitory receptor functions and regulates cell differentiation and activation. It acts as a receptor for both classical and non-classical MHC class I molecules and is a receptor or ligand for the TNF superfamily member TNFRSF14. In NK cells, it can provide a stimulatory signal via PI3K upon ligation of TNFRSF14, enhancing IFNG production and anti-tumor immune response.
